Doing "make dep" creates a flood of "error: invalid
preprocessing directive #asm" messages. Apparently,
preprocessing is done with GCC (options defined in
elks/Makefile-rules)- is there some sort of definition
file that has to be included?

Sorry, nope. #asm is a preprocessor directive, like #pragma or #define.
It just happens to not be one that gpp (GNU PreProcessor) supports
